The Clinical Problem: Tumor Heterogeneity

Many malignant tumors evolve into multiclonal clusters due to high mutation rate and complex interactions with the tumor microenvironment, causing intratumoral heterogeneity that permits treatment resistance, immune escape, and disease progression, culminating in poor patient outcomes.

Activation of the immune system against a breadth of tumor neoantigens in different regions of a tumor will account for intratumoral heterogeneity and may improve patient outcomes:
A. 3DT reaches all tumor regions and activate the immune system against antigens in all regions
B. Current clinical practice of injection with a straight needle reaches limited volume of the tumor

The 3DT Concept

Curved needle contained in a straight needle guide for insertion into the tumor is deployed with different rotation angles and deployment lengths to reach any site within the tumor.

Manual devices enable injection through multiple planar arrays with up to 6 injection sites equally distributed around the needle at radial distances up to 20mm from the needle axis.

The concept of automatic 3DT system is demonstrated by an early feasibility demonstration setup.

A hand-held, mini-robot prototype for autonomous 3DT has been developed  and will be further developed to enable 3DT of tumors with irregular shapes.

Experimental Results

The 3DT devices have been evaluated by investigators at the Center for Interventional Oncology, Radiology and Imaging Sciences, Clinical Center of the NIH.
Results were presented at the 2026 Annual Scientific Meeting of The Society for Interventional Oncology (SIO)  in Savannah, Georgia; and at the 2026 European Conference on Interventional Oncology (ECIO) in Basel, Switzerland.
